Abstract
Objective Fibroblast growth factor 10 (FGF10) signaling pathway is crucial to the reconstruction of lung development and lung epithelial .To investigate the relationship between gene polymorphism and susceptibility in fibroblast growth factor 10 (FGF10 ) to chronic obstructive pulmonary disease (COPD) in Han population of North China. Methods A case–control study of 220 COPD patients and 285 control subject was conducted. Six tagging SNPs in FGF10, including rs10473352, rs16873956, rs2973644, rs1011814, rs10402070, rs723166 and rs723166 were genotyped by RFLP (rs10473352, rs16873956 and rs2973644) and SNaPshot (rs10402070, rs723166 and rs723166) assay. Chi-square test was used to perform the Hardy-Weinberg test. Uncondition logistic of regression model was used to calculate odds ratio (OR) and 95% confidence interval (CI).Age,smoking index, duration of disease,body mass index by t-test. Results The alleles and genotypes of rs2973644 distributed significantly differently in COPD patients and controls (χ2 =6.188,6.021 P< 0.05) The genotypes of rs10473352 distributed significantly differently in COPD patients and controls(χ2 =6.213 P< 0.05). The CC genotype of rs1011814 greatly increased in patients with severer COPD than that in mild COPD. The allele C at rs1011814 was associated with an increased severity of COPD in dominant model analysis (χ2 =6.264 P<0.05; OR: 2.380; 95%CI: 1.193-4.746). Conclusion FGF10 gene SNPsites rs2973644, rs10473352 polymorphism may be associated with susceptibility to COPD in Han population of North China..Thers1011814 SNP may be associated with severity of COPD.
