Summary: 
Important technical advances in lung developmental biology. a) The application of Cre-lox technology and complex fluorescent reporter line to delete genes or overexpress their gene product in specific cell types at targeted times during development, and mark and lineage trace specific cell populations, has dramatically improved our understanding of lung developmental biology in recent decades. b) Advances in three-dimensional culture systems, including differentiation of human lung epithelium from induced pluripotent stem cells (iPSCs) allows experiments to be performed on human lung tissue in ways that were not previously possible. c) Rapid advances in single cell-sequencing technologies have permitted the identification of novel and rare cell subtypes within the lung, shed light on gene-regulatory networks, and allowed for the prediction of lineage relationships, particularly in situations where genetic lineage tracing is not possible. hPSC: human pluripotent stem cell; t-SNE: t-distributed stochastic neighbour embedding.
